miR-339-3p Is a Tumor Suppressor in Melanoma.

چکیده
Determinants of invasion and metastasis in cancer remain of great interest to define. Here, we report the definition of miR-339-3p as a novel tumor suppressive microRNA that blocks melanoma cell invasion without affecting cell survival. miR-339-3p was identified by a comprehensive functional screen of a human miRNA mimetic library in a cell-based assay for invasion by the melanoma cell line A375. miR-339-3p was determined as a strong inhibitor of invasion differentially expressed in melanoma cells and healthy melanocytes. MCL1 was defined as a target for downregulation by miR-339-3p, functioning through direct interaction with the 3' untranslated region of MCL1 mRNA. Blocking miR-339-3p by an antagomiR was sufficient to increase melanoma cell invasion, an effect that could be phenocopied by RNAi-mediated silencing of MCL1. In vivo studies established that miR-339-3p overexpression was sufficient to decrease lung colonization by A375 melanoma cells in NSG mice, relative to control cells. Overall, our results defined miR-339-3p as a melanoma tumor suppressor, the levels of which contributes to invasive aggressiveness. Cancer Res; 76(12); 3562-71. ©2016 AACR.

Long non-coding RNA HOTAIR acts as a competing endogenous RNA to promote malignant melanoma progression by sponging miR-152-3p
HOX transcript antisense RNA (HOTAIR) is associated with the growth and metastasis of many human tumors, but its biological roles in malignant melanoma remain unclear. In this study, we show that HOTAIR is overexpressed in melanoma tissues and cells, especially in metastatic melanoma. High HOTAIR levels correlate with poor prognosis in melanoma patients. We also determined that HOTAIR functions...
متن کاملmiR-339-3p inhibits proliferation and metastasis of colorectal cancer
MicroRNAs (miRNAs) serve important roles in regulating cancer cell proliferation and metastasis. The same hairpin RNA structure may produce mature products from each strand, termed miR-5p and miR-3p, which can bind different mRNAs. Previously, the present authors reported that miR-339-5p could inhibit cell proliferation and migration by targeting the 3'-untranslated region (3'-UTR) of PRL-1 mRN...
متن کاملInhibitory effect of microRNA-605-3p on biological functions of melanoma A375 cells via wnt11
Aims: The aim of the present study is to examine microRNA (miRNA or miR)-605-3p expression in melanoma at tissue and cellular levels, and to understand its biological roles. Methods: A total of 45 melanoma patients (27 males and 18 females) were enrolled in this research. To measure the expression of miR-605-3p, quantitative real-time polymerase chain reaction was employed. Melanoma A375 cells ...
متن کاملRestricted expression of miR-30c-2-3p and miR-30a-3p in Clear Cell Renal Carcinomas enhances HIF2 activity
Author manuscripts have been peer reviewed and accepted for publication but have not yet been edited. Abstract Inactivation of the von-Hippel Lindau (VHL) tumor suppressor gene occurs in 90% of human clear cell renal cell carcinomas (ccRCC), and leads to the stable expression of the hypoxia inducible factors HIF1Į and HIF2Į. The constitutive expression of HIF1Į in a majority of VHL-deficient tu...
متن کاملMiR-142-3p functions as a potential tumor suppressor directly targeting HMGB1 in non-small-cell lung carcinoma.
BACKGROUND microRNAs (miRNAs) play a significant role in cancer development and progression by regulating the expression of oncogenes or tumor suppressor genes. Previous study using microarrays demonstrated that miR-142-3p was downregulated in patients with Non-small-cell lung carcinoma (NSCLC). However, the functional role of miR-142-3p in NSCLC is still unclear. MATERIAL AND METHOD Real-tim...
